Title: A galanin-receptor regulates context-dependent on-food chemotaxis

Abstract : Food searching behavior often involves decision-making processes that can be regulated by contexts. The well-studied chemosensory neural circuit in C. elegans provides an accessible system to address context-dependent modulation of decision-making. We find that on a big lawn of E. coli OP50, the worm still seeks novel foods, such as a small lawn of P. aeruginosa PA14. This on-food chemotaxis is disrupted by mutations in a G-protein coupled receptor npr-9. npr-9 encodes an ortholog of the human galanin-receptors and is expressed in the interneuron AIB1. Expression of npr-9 in AIB is sufficient to rescue the defects in the on-food chemotaxis. Interestingly, the defect of the npr-9 mutants in chemotaxis is specific to the on-food condition, suggesting a role of npr-9 in modulating chemotaxis by context. To characterize the function of npr-9, we analyzed the behavioral effects of mutating tab-1, which encodes a homolog of the mammalian brain-specific homeobox protein that is expressed in a few neurons, including AIB. We find that tab-1 mutant animals are defective in seeking novel foods, similarly as the npr-9 mutants. The mammalian brain-specific homeobox protein interacts with FOXO transcription factors2. We find that FOXO/DAF-16-dependent insulin signaling regulates npr-9 in modulation of chemotaxis. We are currently investigating the mechanisms through which npr-9 and tab-1 regulate the context-dependent decision-making in food chemotaxis. Because tab-1 is involved in harsh touch, we also address how tab-1 regulates two distinct behaviors, on-food chemotaxis and mechanosensation.
